The London Fire Brigade said in a statement that six fire engines and about 40 firefighters were called to the fire on Hamilton Road at around 8.30pm on Thursday.
The fire was extinguished by 9:45 p.m.
Firefighters rescued two women and two children from the first floor of the estate with a nine-meter ladder. All four died at the scene, police confirmed.
The man, who fled the building before firefighters arrived, was taken to hospital.
Andy Roe, London Fire Commissioner, said: “This is a truly horrific incident that is both sad and shocking.
